5-Amino-1MQ is a small-molecule inhibitor of NNMT. It is not a peptide, despite appearing in peptide product catalogs and being discussed alongside peptide therapies. It is typically sold in oral capsule form as a research chemical.
It appears in this formulary because clinicians encounter it in exactly that context — bundled into peptide protocols by online vendors — and need to be able to place it accurately.
What NNMT does
NNMT methylates nicotinamide, a form of vitamin B3. This has two downstream consequences of interest. It consumes methyl groups from S-adenosylmethionine, the body's principal methyl donor, and it diverts nicotinamide away from NAD+ salvage, the pathway that regenerates NAD+ for cellular energy metabolism.
NNMT expression is elevated in the adipose tissue of obese animals and humans. The hypothesis is that inhibiting it should preserve NAD+ availability and raise the metabolic rate of fat tissue. It is a coherent hypothesis grounded in real biochemistry — which is precisely why the compound is marketed persuasively.
What the evidence actually shows
The supporting data are preclinical. In cultured adipocytes and in diet-induced obese mice, NNMT inhibition has been associated with reduced fat mass and favorable metabolic changes without reduced food intake.
Those findings are legitimate laboratory science. They are also several steps removed from a clinical claim. No adequate human clinical trials have established that 5-Amino-1MQ produces fat loss in people, at what dose, with what oral bioavailability, or with what safety profile over any duration.
The gap matters more than usual here. Mouse metabolic studies have an especially poor record of translating to human obesity outcomes, and NNMT sits at the intersection of methylation and NAD+ metabolism — pathways involved in gene regulation and DNA repair throughout the body. Chronic systemic inhibition is not a manipulation whose long-term consequences can be assumed benign without data.
Regulatory status and patient counseling
5-Amino-1MQ is not FDA-approved as a drug and is not an established compounding substance. Products sold online occupy an ambiguous position, sometimes labeled as research chemicals and sometimes marketed as supplements, with the identity, purity and dose accuracy problems common to that market.
The accurate summary for a patient is that the enzyme target is real, the animal data are real, and the human evidence that the product does what its marketing claims does not currently exist.
